An Open-Label Study of Single-Dose Psilocybin for Borderline Personality Disorder With Co-Occurring Major Depressive Disorder

This open-label pilot study (n=9) tested a single dose of psilocybin in adults with borderline personality disorder and major depressive disorder. Depressive symptoms improved over four weeks, but borderline personality disorder symptoms did not change significantly.

The Therapeutic Potential of Psychedelics for Treatment-resistant Depression Reality or Hallucination? A Systematic Review

Systematic review of 15 studies (ten randomised double‑blind controlled trials and five open‑label trials) evaluating typical and atypical psychedelics in treatment‑resistant depression. The evidence indicates psychedelics can produce rapid, clinically meaningful reductions in depressive symptoms across different substance classes, supporting their potential as alternative or adjunctive treatments.

The Relationship Between Participant Pretreatment Clinical Presentation and the Quality of Psilocybin Experience: A Retrospective Analysis

In a retrospective analysis of 233 participants with treatment‑resistant depression given 1, 10 or 25 mg COMP360 psilocybin, dose was the strongest and most consistent predictor of the acute psychedelic experience, while pretreatment clinical characteristics made only modest, variable contributions (positive affect, lower generalized anxiety, higher executive function and greater personality‑disorder symptoms each influenced different experience dimensions). These results challenge the assumption that pretreatment traits are major determinants of the subjective psilocybin experience.

Safety, Tolerability, Pharmacokinetics, and Pharmacodynamics of Subcutaneous RE104: A Double-Blind, Randomized, Single Ascending Dose Placebo-Controlled Study

This double-blind, randomised, placebo-controlled Phase I study (n=48) evaluates the safety, pharmacokinetics, and psychoactive effects of RE104 (psilocybin analog; Luvesilocin; a prodrug of 4-OH-DiPT) in healthy adults with prior psychedelic experience. RE104 was well tolerated up to 40 mg with no serious adverse events, and plasma levels of its active form correlated with subjective drug effect and mystical experience scores. The compound produced psilocybin-like effects with a shorter duration (3-4 hours), supporting further therapeutic investigation.

Effects of a Single Dose of Ayahuasca in College Students With Harmful Alcohol Use: A Single-blind, Feasibility, Proof-of-Concept Trial

In a single-blind proof-of-concept trial in 11 college students with harmful alcohol use, one dose of ayahuasca with psychological support was well tolerated and produced a short-term reduction in days drinking at 2–3 weeks that did not survive correction for multiple comparisons. The study demonstrates feasibility of the protocol and supports larger, controlled trials to assess efficacy.

Interactive Effects of Ayahuasca and Cannabidiol in Social Cognition in Healthy Volunteers: A Pilot, Proof-of-Concept, Feasibility, Randomized-Controlled Trial

In a small randomised, placebo-controlled pilot trial in healthy volunteers, 600 mg oral cannabidiol given before ayahuasca produced no detectable interaction on facial emotion recognition, empathy tasks or subjective effects — both groups showed reduced reaction times and decreases in anxiety/sedation with no between-group differences. Ayahuasca with or without CBD was generally well tolerated, supporting feasibility of further, larger trials to confirm these null-interaction findings.
